De acordo com o Instituto Nacional de Saúde Doutor Ricardo Jorge (INSA),  que é o laboratório de referência nacional, o teste utilizado para diagnóstico laboratorial à covid-19 é a pesquisa molecular de SARS-CoV-2 por RT-PCR (reverse transcriptase - polymerase chain reaction), pesquisa do ácido nucleico com uma reação de PCR em tempo real.

Em circular normativa conjunta, a Direção-Geral da Saúde (DGS), o Instituto Nacional de Saúde Doutor Ricardo Jorge (INSA) e a Autoridade Nacional do Medicamento e Produtos de Saúde (Infarmed) esclarecem que "o diagnóstico de novos casos de infeção por SARS-CoV-2 deve ser realizado exclusivamente por testes de deteção de componentes do vírus, sendo os testes de biologia molecular [RT-PCR] para a deteção de RNA considerados de referência, por apresentarem maior fiabilidade".
